Mechanism of action
SR-17018 is a small-molecule G-protein-biased μ-opioid receptor (MOR) agonist developed as a research probe for biased signal transduction at opioid receptors. It preferentially activates G-protein-mediated signaling over β-arrestin recruitment, a profile associated in preclinical models with robust analgesia alongside reduced respiratory depression and attenuated tolerance liability.
Receptor pharmacology
In vitro binding studies report Ki values of approximately 11 nM at the μ-opioid receptor (MOR), 68 nM at the κ-opioid receptor (KOR), and >10,000 nM at the δ-opioid receptor (DOR), indicating strong MOR selectivity over DOR and modest KOR affinity.
Bias factor
Functional assays quantify a signaling bias factor of roughly 80–100 relative to the reference agonist DAMGO, reflecting preferential G-protein pathway activation with diminished β-arrestin2 recruitment.
Preclinical analgesia profile
Animal models demonstrate dose-dependent antinociception with a wider therapeutic window for respiratory effects compared with morphine, and reduced antinociceptive tolerance on repeated dosing.
